A GPC chromatograph is a liquid-phase device similar to an HPLC unit. It separates the components of a mixture for analytic purposes based on molecule size (size exclusion). The mobile phase, the eluent, carries the solution containing the mixture through the column. Molecules of varying size take different amounts of time to pass through. A detector analyzes them as they leave the column.
ApplicationsThese devices are used primarily to separate macromolecules for molar mass measurement.
TechnologiesAn aqueous liquid constitutes the mobile phase. The stationary phase consists of porous polymer beads, a gel, which captures the smallest molecules.
There are two types of column calibration. Universal calibration uses columns containing a homogeneous porous material. Linear calibration employs heterogeneous porous material which traps molecules of varying size.
The specific application will determine choice of calibration system, using a stationary phase either of polymer gel or of silica gel. Another important element is whether the chromatograph is equipped with a refractive index detector or an ultraviolet detector.
