OverviewThe PPSQ is an automated system for determining N-terminal amino acid sequences of proteins and peptides by combining an Edman reaction module with high-performance liquid chromatography (HPLC). Two model variants are available: PPSQ-51A (single reactor) and PPSQ-53A (three reactors). The PPSQ-53A enables continuous sequential analysis of multiple samples.
Benefits of using Edman degradation for amino acid sequencing- Direct determination of the N-terminal sequence
- No sample pre-treatment required in many cases
- Ability to differentiate isobaric residues (leucine vs isoleucine)
- Can identify proteins not present in sequence databases
Key features (summary)- Automated Edman degradation with online derivatization to produce stable PTH-amino acids
- Two models: PPSQ-51A (1 reactor) and PPSQ-53A (3 reactors) for single or continuous multi-sample workflows
- Online injection of PTH-amino acids into an HPLC system for separation and detection
- Support for isocratic and gradient HPLC modes; gradient option includes a high-sensitivity flow cell for trace analysis
- LabSolutions PPSQ software for chromatogram processing and amino acid sequence estimation; supports FDA 21 CFR Part 11 features
Features (detailed)- Edman reaction module: stepwise N-terminal cleavage and derivatization producing PTH-amino acids for reliable identification
- Isocratic system: separates PTH-amino acids isocratically to improve baseline stability and reproducibility for routine analyses
- Gradient system: high-sensitivity flow cell and gradient elution for trace-level PTH-amino acid detection and sequential analysis of low-abundance samples
- Multi-reactor configuration (PPSQ-53A): enables queued, continuous processing of multiple samples to increase throughput
- Data handling: HPLC chromatograms are saved to PC; LabSolutions PPSQ software performs chromatogram processing, peak identification and sequence estimation with audit trail and user management options
